3. Antifungal Azole Panel by 2D-LC-MS/MS

A simple high-throughput LC-MS/MS method was developed to quantify fluconazole [FLU], voriconazole [VOR], posaconazole [POS], itraconazole [ITR] and hydroxy-itraconazole [OH-ITR] in human serum. Protein precipitation was used as sample pretreatment with stable isotope-labeled internal standard. 2D LC allows short run time (4 min) and secures specificity and robustness. The method was compared to different methods and further validated by participating in external quality assurance program. The method was linear over the range of 0.05 to 10.00 mg/L for POS, 0.10 to 20.00 mg/L for VOR, ITR and OH-ITR, 1 to 200 mg/L for FLU, sufficient for therapeutic drug monitoring.
