An LC-MS/MS method for the endocannabinoids arachidonoyl ethanolamide (AEA) and 2-arachidonoyl glycerol (2AG) together with an easy sample preparation technique that involves liquid extraction from biological matrices with toluene is presented. Chromatographic separation of 2AG and 1AG allows for monitoring the auto-isomerization process of 2AG. The use of toluene for liquid extraction provides two important benefits: 1. Isomerization from 2AG to 1AG is minimized during the evaporation of toluene as compared to protic organic solvents such as methanol or ethanol. 2. Toluene extracts contain less phospholipids that contribute to the matrix effect possibly decreasing sensitivity of the analytical method.
