37. A Fully Automated Turbulent Flow Liquid Chromatography Isotope Dilution Mass Spectrometry (LC-IDMS) Method for Quantitation of Voriconazole in Human Serum

We demonstrate a fully automated isotope dilution mass spectrometry (IDMS) traceable LC-MS/MS method using direct serum injection with automated online solid phase extraction (SPE). Internal standard (Voriconazole D-3) was added in a 1:1 dilution via automated injection by autosampler. Separation of drug from matrix was achieved by using a TurboFlow® C18-XL column. Chromatographic separation was achieved using a Hypersil Gold® (C18) column. The analytes were ionized by HESI probe and ion transitions were 350.1 m/z and 281.0 m/z for Voriconazole and 353.17 m/z and 284.0 m/z for Voriconazole D-3. Total run time for SPE and chromatography was under 8 minutes.
