43. Fast and Sensitive Amino Acid Analysis using aTRAQ Derivatization and UPLC-MS/MS

HPLC-MS/MS analyses have greatly increased sample throughput, decreased LLOQs and increased selectivity for amino acid analysis (AAA). Additionally, specially developed amine reactive isotope coded affinity tag (aTRAQ™) reagents create unique internal standards for each amino acid allowing for more accurate and robust quantitation. Ultra high performance liquid chromatography (UPLC) allows one to shorten analysis time while achieving the highest sensitivity. In this report, the authors will show the results of combining an AB Sciex QTRAP mass spectrometer, with aTRAQ™ sample derivatization, to UPLC where limits of detection in the 5-50 nM range were achieved in under 10 minutes.
