Objective was to study the applicability of an immobilized liquid in a rapid sample preparation for quantification of four immunosuppressants (Cyclosporin A, Everolimus, Sirolimus and Tacrolimus) by LC-MS/MS. The immunosuppressants were extracted from 20µL blood samples and blood calibration standards using an immobilized liquid, e.g., coated on a porous material stuffed into a pipette tip. The extracts were analyzed by LC-MS/MS and results compared with analyses based on protein precipitation followed by OnLine-SPE LC-MS/MS. The developed method using an immobilized liquid was suitable to reproducible extract the immunosuppressants in less than 3 minutes, supporting sequential quantitative analysis by LC-MS/MS. |
