33. UPLC-MS/MS Measurements of 25(OH) Vitamin D2 and 25(OH) Vitamin D3 in the Pediatric Population in Atlanta, GA at Latitude 33° North

Vitamin D has long been recognized as an important nutrient in the pediatric population for the prevention of rickets. We optimized a UPLC-MS/MS assay for the measurement of 25(OH)D2 and 25(OH)D3 in the blood of children in our pediatric hospital. The method uses a liquid-liquid extraction using hexane. We performed these assays on a Waters Acquity UPLC with an Acquity TQ Detector equipped with a BEH Phenyl column. The assay proved linear from 3.5-200ng/mL, and the precision ranged from 7.3-11.0% for 25(OH)D2 and 6.0-7.4% for 25(OH)D3. We assayed 300 samples from children ages 1-18 years between June–August.
