32. Intact and Functional Mitochondria from Solid Tissue: Effective Semi-Automated Extraction Using Gentle Mechanical Homogenization and Pressure Cycling Technology

Proteomic and metabolomic profiling of tissue mitochondria can provide important insights into mitochondrial function and dysfunction, and may address fundamental questions of cell energetics and oxidative stress. Here we describe a semi-automated method utilizing a gentle mechanical homogenizer (The Shredder SG3) and hydrostatic pressure cycling to release mitochondria from tissue with minimal hands-on time. Our data demonstrate that mitochondria extracted by the PCT sample preparation system are intact and functional. LC-MS/MS mass spectrometry confirms that the overall protein pattern of mitochondria extracted with The Shredder SG3 and pressure cycling is comparable to control samples isolated using a conventional manual homogenizer.
