Proteomic and metabolomic profiling of tissue mitochondria can provide important insights into mitochondrial function and dysfunction, and may address fundamental questions of cell energetics and oxidative stress. Here we describe a semi-automated method utilizing a gentle mechanical homogenizer (The Shredder SG3) and hydrostatic pressure cycling to release mitochondria from tissue with minimal hands-on time. Our data demonstrate that mitochondria extracted by the PCT sample preparation system are intact and functional. LC-MS/MS mass spectrometry confirms that the overall protein pattern of mitochondria extracted with The Shredder SG3 and pressure cycling is comparable to control samples isolated using a conventional manual homogenizer. |
