40. LC-MS/MS Method for the Measurement of Serum Mevalonic Acid: A Novel Marker of HMG-CoA Reductase Inhibition by Statins

Mevalonic acid (MVA) is an early precursor in cholesterol biosynthesis and a useful measure of statin efficacy. A specific and sensitive liquid chromatography-tandem mass spectrometry method for measuring serum MVA has been developed using solid-phase extraction and a Luna PFP reversed-phase chromatography system. Analysis was performed using an API 5000 tandem mass spectrometer. The method showed excellent recoveries (94% ± 7) and imprecision (intra-assay CV 8.6% [3 ng/ml] and 4.9% [6 ng/ml], inter-assay CV 15.5% [6 ng/ml]), with a calibration range up to 50 ng/ml and limit of detection at 0.1 ng/ml, for clinical use in healthy and statin-treated individuals.
