61. Toxicological Screening of 80 Drugs in Urine Samples Using the High Resolution Exactive LC/MS Orbitrap Mass Spectrometer Coupled to Online Extraction and Turbulent Flow Chromatography

A highly specific and sensitive assay using TurboFlow chromatography coupled to orbitrap-based LC-MS system in urine was developed and validated to monitor patient compliance and to assess the use of additional legal and illegal drugs in pain clinics. Using the Orbitrap mass spectrometer (Exactive, ThermoFisher) in connection with HPLC and TurboFlow chromatography, a mixture of 80 drugs was separated and detected within 13 minutes. The lower limit of detection for most drugs was <10 ng/mL. One advantage of the high-resolution orbitrap in comparison to triple-stage quadrupole mass spectrometry is that always full scan high-resolution spectra are recorded allowing for non-targeted database searches.
