71. Determination of Conjugated and Unconjugated Tapentadol (Nucynta®) and N-Desmethyltapentadol in Authentic Urine Specimens by UPLC/MS/MS

A total of 51 urine specimens from pain management patients dosed with Nucynta® (Tapentadol) were analyzed for the presence of tapentadol, tapentadol glucuronide, N-desmethyltapentadol and N-desmethyltapentadol glucuronide using Ultra Performance Liquid Chromatography Tandem Mass Spectrometry (UPLC/MS/MS) in multiple reaction monitoring (MRM) mode. An initial investigation into the characterization of metabolites was performed using a hybrid quadrupole-time of flight (QTOF) mass spectrometer. Data using accurate mass determination and structural elucidation confirmed the presence of predicted metabolites and aided the MRM method development. The mean relative percentages of tapentadol, tapentadol-glucuronide, N-desmethyltapentadol and N-desmethyltapentadol glucuronide were 6%, 89%, 1% and 4%, respectively.
