77. Urine and Saliva Cortisol Measurement by LC-MS/MS

The aim of this study was to develop a liquid chromatography tandem mass spectrometry method for the measurement of cortisol in saliva and urine. Samples were prepared by protein precipitation with methanol and separation on a C8 column after which measurement was performed on a triple quadrupole system equipped with an electrospray ionization source. For both urine and saliva cortisol, slope and intercept, calculated using Passing and Bablok regression analysis, was not significant (P<0.05) when compared to reference laboratory, coefficients of determination was 0.98 and 0.96 respectively. A possible advantage of this method is a straight forward sample preparation step.
