74. Validation of Immunosuppressant Calibrator and QC Reagents Using On-Line Extraction Coupled to LC-MS/MS

We implemented an immunosuppressant method for measuring cyclosporine A, sirolimus and tacrolimus in whole-blood using an Aria TLX2-TurboFlow™ system with on-line extraction and atmospheric pressure chemical ionization tandem mass spectrometry (TLX-APCI-MS/MS). Our goal was to compare the accuracy and precision of two whole-blood calibrators/control systems: Thermo Scientific ClinSpec™ Immunosuppressants Test and ChromSystems 6PLUS1(R) Multilevel Calibrator Set/MassCheck(R) reagents.
Overall we found both reagents systems demonstrated similar linearity and accuracy. An advantage of utilizing the ClinSpec™ reagent system is lower specimen volumes are required. We also found that the ClinSpec™ control reagents were gave better precision for cyclosporin A and sirolimus measurements.
