57. Detection of Age-Related Histone PTMs as M/Z Pair Tags in LC-MS Measurement of Mesenchymal Stem Cell Lysates

Post Translational Modifications (PTMs) are important in protein signaling cascades and control mechanisms. We explore histone aging behavior along with associated PTMs by analyzing young and old mesenchymal human stem cell lysates using the BRIMS two-pass workflow. The first pass focused upon optimized full scan accurate mass for robust quantitation. These data are processed with a novel algorithm to tag modified and unmodified candidates using predicted accurate mass differences of expected PTMS. An inclusion list of these candidates is used for a second LC-MS acquisition optimized for ms/ms. The spectra are then analyzed with Mascot and SEQUEST.
