77. A High-throughput LC-MS Assay for Drugs of Abuse in Urine

We have developed an automated quantitative LC-MS/MS drug screening method for 44 compounds (including amphetamines, benzodiazepines, cocaine metabolite, opiates and cannabinoids) with a daily capacity of >500 samples. Non-hydrolyzed urine samples were diluted 1:10 in an albumin solution containing 22 internal standards and analyzed by LC-MS/MS using 160 MRMs. Excel®-based middleware was used to both generate sample lists and process LC-MS data to generate simple ‘pos/neg’ results for each drug group. Qualifier ion ratios, retention times and the presence of related compounds expected to be present (e.g. 6-MAM / morphine) were the key indicators used for results verification.
