The present work describes developments using DESI-MS for the examination of intact tissue sections. DESI-MS imaging has been increasingly applied to the analysis of human cancer tissue with the goal of providing biochemical information for clinical diagnosis. The technique has been applied to the study of human brain cancers by examining fresh tissue fragments removed by a stereotaxic ultrasonic surgical device, and thin tissue sections of human gliomas. In both cases, the lipid profiles correlated well with the histopathological examination and in the latter case, provided information toward the differentiation of grade within the human astrocytoma.
