Evaluation of Multiplexing Capabilities Using Peptide Immunoaffinity Enrichment and Multiple Reaction Monitoring Mass Spectrometry for Quantitation of Peptides

There is a great need for quantitative assays in proteomics. In SISCAPA (stable isotope standards and capture by anti-peptide antibodies), a peptide is quantified as a stoichiometric representative of the protein from which it is cleaved. Advantages of using SISCAPA include its sensitivity, specificity, and affordability. In this presentation, we sought to advance the utility of SISCAPA by testing the performance of multiplexing up to 50 peptide analytes into one assay. Eighty-five percent of the analytes showed super-imposable results at all plex levels indicating it is easily possible to reach a high multiplexing level (≥50-plex).
