A Comparison of Immobilized Liquids Used for Sample Clean-up Prior to LC/MS Analysis of Antidepressants and Active Metabolites

A novel technique has been developed using immobilized liquids for extraction of nine basic antidepressants (e.g., venlafaxine, citalopram, fluoxetine, duloxetine) and two active metabolites (O-desmethylvenlafaxine and norfluoxetine) in plasma by LC-MS/MS. Immobilized liquids were chosen as the extraction media as they offer reduced solvent usage, reduced matrix effects and the potential for automated sample preparation. Two different proprietary Immobilized liquids were coated on a high surface area porous material inserted into pipette tips which were subsequently used to extract the target analytes. A medium polarity immobilized liquid and an immobilized ion-exchange liquid was compared to a SPE method.
