When trying to quantify an endogenous protein, why would one calibrate with a synthetic peptide? Does it matter? Why even calibrate? In this presentation, the pros and cons will be discussed regarding the continuum of options for both bottom-up (digestion) and top-down (intact) “absolute” protein quantification.
Topics will include discussion on 1) the difference between calibrators and internal standards, 2) the nature and hierarchy of protein calibration, 3) authenticating purified protein and peptide standard materials, 4) modes of protein calibration, and 5) qualification of viable calibrator matrices.
